Development of strain-specific SCAR primers to beneficial rhizobacteria Bacillus subtilis OSU-142

Background and purpose: Synthetic fertilizers damage the environment. Biofertilizers that consist of microorganisms emerge as an environmentally friendly alternative. improve plant growth by mobilizing soil nutrients, triggering hormone synthesis, or competing with pathogenic bacteria. However, biofertilizers often fail due to insufficient colonization roots.Materials methods: To explore dynamics a bacterial strain commonly used in biofertilizers, Bacillus subtilis OSU-142 (OSU-142), developing set primers specific was aimed. Since its genome is unknown, identify genomic regions unique strain, DNAs more than 40 strains were fingerprinted, most which belong using Randomly Amplified Polymorphic DNA (RAPD) method.Results: This approach identified polymorphic band at 880 bp, then cloned sequenced. The sequence showed no perfect match any known sequences tested databases indicating region highly unique. primer converted Sequence-Characterized Region (SCAR) primers, functionality detecting confirmed. Newly designed can specifically detect OSU-142.Conclusions: These be useful for basic science commercial applications on tracking various environments, thus contributing biofertilizers’ adoption long run.
